I am looking to buy an English Mastiff and I want to know the best way, preferably on the internet, to find reputable breeders with available or upcoming litters.  I live in San Diego, and would be willing to travel within a few hours to find a Mastiff.  Is there anything that I should know about securing a puppy from an upcoming or currently available litter?

A lot of people sell their dogs on the internet, doesn't make them reputable just visible.  You can scan a plethora of websites but seeing is believing.

It's important to see both parents, and if possible any relatives and the condition they are raised in.  With the large breeds, you want a line with good hips and a clean bill of health.  Enlarged hearts, heart mummers and temperament are something you can't pick up by looking at a picture.   

Personally, I look for a breeder (regardless of the breed), who keeps something back to continue the lines, I believe those are the breeders that are trying to strengthen their breed of choice as just taking 2 dogs breeding them and selling the pups to recoup their finances.

A written contract is important and a must.

I would start with organizations like the Mastiff Club of America, the CKC, and may read a few Mastiff Message Boards.  Ask questions about breeders, find out the number of complaints and find out what others are saying about them and their dogs.
