You can generally tell a difference in their behavior. A lot of times they will start building a nest and they will get a little bit more tired quickly. They will sometimes become more aggressive and will also balloon up on their tummy. Generally you don't know that they are really pregnant until they are a few days from giving birth. Hamsters stay pregnant for about 16-20 days.
