Question
If two cats have been living together for two years, then suddenly seperated, how will they react. Will they grieve for one another?

Hello!

This totally depends on how much they liked each other in the first place. Cat behaviour can be very different, sometimes two cats living in the same house are in very good terms, they eat together, sleep together cuddling together, never fight, etc. If this is the case, they will probably grieve for each other if separated, and you may even experience some severe behavioural changes.
If they are two cats who never got along in the first place, always fighting, or totally indifferent with each other, then I don't think separating will be a problem, it may in fact change the behaviour of aggressive cats and help them become nicer cats.
