The easiest way to tell if your guinea pig is pregnant is to take her to the vet and ask.  :)  However, if you want to check on your own, you will need to "palpate" her.  This means that you will feel her stomach for a tell-tale sign of pregnancy.

Keep in mind that you usually cannot feel these signs until at least 3-4 weeks along, sometimes longer, so it isn't 100% reliable, and that if you have any questions about what you figure out on your own, it is always easiest to ask a vet.

To palpate a guinea pig, you will place her gently onto her back with her head facing away from you.  With the fingers of one hand, you will gently feel her lower abdomen area, near her legs.  What you are feeling for is two hard, egg-shaped lumps.  If you feel these, she is pregnant, and you can expect a nice litter of pups.  If not, you may want to wait a week or so and then try again or take her to the vet.
